HPLC

Also called high-performance liquid chromatography · reverse-phase HPLC · RP-HPLC

HPLC (high-performance liquid chromatography) separates compounds by their affinity for a stationary phase as a mobile phase carries them through. For peptides it is the standard method for measuring purity — the area of the target peak as a percentage of total peak area gives the purity figure (e.g. ≥98%). A chromatogram trace on a COA is the primary evidence of purity.
